I am working on the GABA metabolism in Arabidopsis thaliana. GABA, the main inhibitory neurotransmitter in mammals, is accumulating in plants under various stress conditions. We have isolated T-DNA insertion lines for all genes known to be involved in the GABA-shunt. Plants with a knock out of one gene involved in GABA catabolism (ssadh) display a severe growth phenotype. Since this growth phenotype could be rescued by shutting of the pathway earlier, an EMS mutagenesis was performed on ssadh-mutant seeds. In the M2 generation, plants with improved growth were isolated and the EMS-induced mutation shall be localized by mapping using SSLP-markers. Additionally, a possible influence of GABA (endogenous or exogenous) on plant growth and development shall be figured out.
